Background: NKp46/NCR1

NKp46, also called NCR1, along with NKp30 and NKp44, are activating receptors that are collectively known as the natural cytotoxicity receptor (NCR) family (1). NKp46 is a type I transmembrane glycoprotein with two extracellular Ig-like domains followed by a short stalk region in the extracellular domain (ECD), a transmembrane domain containing a positively charged amino acid residue, and a short cytoplasmic tail. The mature ECD of human NKp46 shares 61% and 63% amino acid sequence identity with mouse and rat NKp46, respectively. NKp46 is unique among the NCRs in its evolutionary conservation between species (2). NCRs are expressed almost exclusively by NK cells and play a major role in triggering some of the key lytic activities of NK cells. Through its positive charge in the transmembrane domain, NKp46 associates with the ITAM-bearing signal adapter proteins, CD3 zeta and Fc epsilon R1 gamma (3, 4). NKp46 is involved with tumor cell recognition and has the capacity to bind to viral hemagglutinins, such as that of influenza and Sendai viruses (5, 6). NKp46 is also crucial for the development of type 1 diabetes, the clearance of Streptococcus pneumoniae, killing stellate cells in attenuated liver fibrosis, and the regulation of graft-versus-host disease and allergic response (7-10).
