Profile Cytokines Secreted by Helper T Cells and Regulatory T Cells
CD4+ T cells are subclassified into helper T (Th) cells and regulatory T cells based on their cytokine repertoire2. Although Th1 secreted cytokines are associated with proinflammatory and antitumor responses, Th2 secreted cytokines are correlated with anti-inflammatory responses. Th17 responses play a role in autoimmunity and allergic responses. Finally, Tregs suppress deleterious Th cell activity. It is no surprise that cytokines secreted by Tregs are associated with immunosuppression and cancer progression.

To address this issue, 12 Italian healthcare centers collaborated to understand vaccine efficacy in a large transdisease cohort that was excluded from vaccine trials. In this peer-reviewed Science Advances paper, investigators used Quantikine™ ELISAs and Proteome Profiler™ Antibody Arrays to understand the relationship between a novel biomarker candidate, acetylated 676th lysine transforming growth factor–beta-induced protein (TGFBIp K676Ac), the cytokine storm, and COVID-19 severity. In this Nature paper, investigators used Simple Plex immunoassays and the R&D Systems Th9/Th17/Th 22 Discovery Performance Assay to profile cytokines in patients with COVID-19 and dengue haemorrhagic fever (DHF). Their data indicate that these patients have a complex cytokine milieu that varies as a function of disease type and progression. Download the publication to learn more.
SARS-CoV-2 pathophysiology is incompletely understood. To address this issue, De Biasi et al., examined the T cell compartment in severe COVID-19 patients. The authors used the Human Cytokine Discovery Panel to profile 31 cytokines in severe COVID-19 patients. Their data highlighted, among other things, the importance of IL-17 in severe COVID-19 patients.
Individuals with lymphoid malignancies (LM) were not included in SARS-CoV-2 mRNA vaccine clinical trials. Since vaccines have reduced efficacy in LM patients, Marasco et al., wanted to evaluate the immune response to the novel SARs-CoV-2 mRNA vaccines. Investigators do not have a complete understanding of the risks and benefits of SARS-CoV-2 vaccination for solid organ transplant patients.
