Background: Caveolin-1

Caveolin-1 (CAV1) belongs to caveolin family which encodes integral membrane proteins of 21kD-24 kD size and forms structural component of caveolar membrane in caveolae, the specialized domains of plasma membrane implicated in sequestration of lipids/proteins. Caveolin-1 as well as caveolin-2 are abundantly expressed in fibroblasts, differentiated adipocytes, smooth/skeletal muscle and in endothelial cells, whereas, caveolin-3 expression is limited to muscular tissues. Caveolin 1 interacts with a broad range of proteins that includes GLIPR2, NOSTRIN, SNAP25, syntaxin, rotavirus A NSP4, DPP4, CTNNB1, CDH1, JUP, BMX and BTK. It interacts directly with G-protein's alpha subunits for regulating their activity and is also implicated in costimulatory signal essential for TCR-mediated activation of T-cells. Caveolin 1 binding to DPP4 induces T-cell proliferation as well as activation of NFkB in TCR/CD3-dependent manner and also recruits CTNNB1 to caveolar membranes for regulating CTNNB1-mediated signal transduction via WNT pathway. Defects in caveolin-1 have been linked to congenital generalized lipodystrophy type 3 (CGL3), and it is also implicated in cardiovascular/pulmonary diseases and its role in carcinogensis is context/cancer dependent, wherein it suppresses genes that favor tumor development, and at the same time it promotes focal adhesion turnover as well as migration of metastatic cancer cells.
